Cryptotanshinone Inhibits Ovarian Tumor Growth and Metastasis by Degrading c-Myc and Attenuating the FAK Pathway

"Cryptotanshinone (CT), a natural compound derived from Salvia miltiorrhiza Bunge (Danshen, a traditional Chinese medicine), has been shown antitumor activity in various cancers. However, it remains unclear whether CT has a potential therapeutic benefit against ovarian cancers. The aim of this study was to test the efficacy of CT in ovarian cancer cells in vitro and using orthotopically implanted HEY A8 human ovarian cancer cells into NSG mice to determine its antitumor efficacy and explore the molecular mechanism underlying CT’s antitumor effects. Results We found that CT inhibited the proliferation, migration, and invasion of OVCAR3 and HEY A8 cell while sensitizing the cell responses to chemotherapy drugs paclitaxel and cisplatin. CT also suppressed ovarian tumor growth and metastasis in immunocompromised mice orthotopically inoculated with HEY A8 cells. Mechanistically, CT degraded oncogene c-Myc by promoting ubiquitination and disrupting the interaction with its partner protein Max. CT also attenuated the FAK (focal adhesion kinase) signaling and degraded FAK in both cell lines. Knockdown of c-Myc using lentiviral CRISPR/Cas9 nickase resulted in reduction of FAK expression, which phenocopies the effects of CT and 10058-F4. Conclusions Our studies demonstrate that CT inhibits primary ovarian tumor growth and metastasis by degrading c-Myc and attenuating the FAK pathway."

@daniel great, it seems to be a very useful anticancer compound, for example in combination with Baicalein and Andrographis.

@daniel I'm not familiar with liposomal products, can I open the caps and toss the content in a smoothy?

@j we can always do that, Johan, but for max absorption it's best to take liposomal formulations on empty stomach.

I wasn't aware apigenin was good for sleep (noticed this effect after taking 1 capsule last night ).

Getting sleep right (Andrew D. Huberman, Ph.D.):

1. Sunlight exposure w/in 30m of waking for 10-30m

2. Avoid bright light (all colors) 10pm-4am

3. *If safe for you*: Mg Threonate (~140mg), Theanine (100-300mg), Apigenin (50mg) 30-45m prior to sleep

4. Keep the room cool

5. No caffeine post 2pm

6. No alcohol

@j Johan, it's outstanding in that area. For all new introduced products, we generate supporting slides for partner clinics (that we are not allowed to share publicly so that they are not seen as health claims) indicating the major areas of potential benefit for each extract. Apigenin comes out from literature as very relevant in the "good for sleep" category, and it has been highlighted in our slides. Very nice to know you already feel the positive impact of that.

Btw, we will have by the end of this year a supplement with a huge amount of AKBA. The one we have now already has the highest amount in the World. The new one will probably have 6x more - designed for clinical trials - we will probably make it available only via clinics/doctors. But I will send you one to try when available.
